Its chirality makes the adverse effects more complicated. This study aimed at the enantioselective cardiotoxicity in developmental zebrafish induced by racemic (rac-)/S-/R-fluoxetine. The accumulation profiles demonstrated that biotransformation of fluoxetine to norfluoxetine occurred during rac-fluoxetine exposure, with a higher enrichment of S-norfluoxetine than R-norfluoxetine. Heart malformations including pericardial edema, circulation abnormalities, and thrombosis were observed, and enantioselective changes also occurred. According to H&E staining and Masson’s trichrome staining, the loose severity of cardiac structure and cardiac fibrosis in rac-norfluoxetine treated group was worse than that in fluoxetine treated groups. Results of toxicity-associated parameters in our homochiral enantiomers’ exposure also indicated that the toxicity induced by S-fluoxetine was more severe than R-fluoxetine. Due to their massive use worldwide, pesticides represent a latent risk to both humans and the environment. In the present study, 1056 frequently used pesticides were screened for oestrogen receptor (ER) agonistic activity by using in silico methods. We found that 72 and 47 pesticides potentially have ER agonistic activity by the machine learning methods random forest (RF) and deep neural network (DNN), respectively. Among endocrine-disrupting chemicals (EDCs), 14 have been reported as EDCs or ER agonists by previous studies. We selected 3 reported and 7 previously unreported pesticides from 76 potential ER agonists to further assess ERα agonistic activity. All 10 selected pesticides exhibited ERα agonistic activity in human cells or zebrafish. In the dual-luciferase reporter gene assays, six pesticides exhibited ERα agonistic activity. Additionally, nine pesticides could induce mRNA expression of the pS2 and NRF1 genes in MCF-7 cells, and seven pesticides could induce mRNA expression of the vtg1 and vtg2 genes in zebrafish. Importantly, the remaining 48 out of 76 potential ER agonists, none of which have previously been reported to have endocrine-disrupting effects or oestrogenic activity, should be of great concern. This study conducted a nationwide monitoring of microplastic pollution along the Korean coasts using filter-feeding bivalves (including oyster, mussel, and Manila clam) as bioindicators to identify the national contamination level and characteristics of microplastics. Seawater sample was collected from the same sampling stations of oyster and mussel for comparison. Microplastics were widely distributed in both coastal bivalves and waters with mean concentrations of 0.33 ± 0.23 n/g (1.21 ± 0.68 n/individual) in oyster/mussel, 0.43 ± 0.32 n/g (2.19 ± 1.20 n/individual) in Manila clam, and 1400 ± 560 n/m3 in seawater. Despite the lack of significant relationship in the abundance of microplastics, their dominant features such as size, shape, color and polymer type were similar between bivalves and seawater. Fragments (69% for oyster/mussel, 72% for Manila clam, and 77% for seawater), particles smaller than 300 μm (96% for oyster/mussel, 83% for Manila clam, and 84% for seawater) and colorless (79% for oyster/mussel, 85% for Manila clam, 75% for seawater) were the dominant shape, size and color, respectively. The major polymer types were polypropylene, polyethylene, and polyester. The microplastic level in bivalves was relatively high in urbanized areas with a wide diversity of polymer types compared with those in non-urbanized areas, and the proportion of polystyrene in the Korean samples was abundant compared with other regions due to wide use of polystyrene products in Korea. Our result suggests that microplastic contamination is widespread in the Korean coastal environment, and bivalves can reflect the microplastic pollution characteristics of the surrounding waters where they live.The European Union (EU) Water Framework Directive (WFD) designates as “high status” rivers, lakes, transitional and coastal waters that are close to natural status and relatively un-impacted by anthropogenic activities. These high status water-bodies (HSWs) are sensitive areas that require special attention. Ireland had a globally important distribution of HSWs (10.5% of rivers and 16.2% of lakes classified as high ecological status in Europe occurred in Ireland), but there have been declines of almost 50% between 1987 and 2018, with excessive sediment implicated as a pressure. In this study, an extensive assessment of macro-invertebrate sediment metrics were used to assess sediment as a pressure in sixty-five high or formerly high status river sites in Ireland that were determined to have either “Lost” their high status (e.g. gone from high to good, moderate, poor or bad; 20 sites); consistently “Maintained” high status (24 sites); or “Gained” in status (e.g. from good to high; 21 sites). Symptom misattribution is a central process in the nocebo effect but it is not accurately assessed in current side effect measures. We have developed a new measure, the Side Effect Attribution Scale (SEAS), which examines the degree to which people believe their symptoms are treatment side effects.
The SEAS was tested in three New Zealand studies a vaccination sample (n=225), patients with gout or rheumatoid arthritis (n=102), and patients switching to a generic medicine (n=69). The internal reliability of the scale was examined using Cronbach’s alpha. To assess validity, the Side Effect Attribution Total Score and Side Effect Attribution Binary Score were related to a number of psychological measures associated with side effect reporting.
The scale showed good internal reliability across the three studies, with Cronbach alphas ranging from 0.840 to 0.943. Analysis of the effect sizes showed that the Attribution Total Score was generally more strongly associated with nocebo responding than Attribution Binary Score.
