Thus, herein, we conducted an immunohistochemical prostasin study in 119 resected OSCC cases. Prostasin expression was decreased in 63% (75/119) of cases. OSCC with decreased prostasin immunoreactivity (low prostasin cases) tended to show a higher histological grade (p = 0.0088) and a more infiltrative cancer cell morphology (p = 0.0024). We then explored the role of prostasin in the OSCC cell lines SAS and HSC-4. SAS did not express detectable prostasin levels, whereas HSC-4 expressed low but distinct levels. Prostasin overexpression suppressed the proliferation and migration of both OSCC lines in vitro. Conversely, prostasin silencing significantly enhanced growth rates of HSC-4. Finally, we analyzed the impact of prostasin expression on the prognosis of patients with OSCC; decreased expression tended to correlate with shorter overall survival (p = 0.0291) after resection. This trend was supported by our analyses using a public database (Kaplan-Meier plotter) of head and neck squamous cell carcinomas. However, the demand for milk and milk-related products far exceeds the supply. The objective of the study was to evaluate dairy farm-workers’ knowledge of factors responsible for culling and mortality of dairy cows in the Eastern Cape Province. Data was collected from 106 dairy farm-workers using a questionnaire. Any correctly answered question by the majority amounted to a point and a zero for incorrectly answered questions. Correct answering by the majority to more than half the questions of a subsection amounted to a pass. A less than 50% pass rate was considered a poor level of knowledge, 51-69% pass rate was considered an average level of knowledge, and anything higher than that was considered a good level of knowledge. Most farm-workers (66.0%) relied on their colleagues for dairy health information. Most dairy farm-workers (49.1%) indicated that lameness, milk fever (56.6%), and mastitis (47.2%) do not lead to culling and mortality of dairy cows. A majority (83%) of farm-workers agreed that reproduction problems, poor milk yield (77.3%), and age (81.1%) are the main reasons for culling dairy cows. The participants had varying perceptions and limited knowledge (28.3%) about the major contributing factors of culling and mortality. The lack of training courses and minimal use of other sources of information such as the internet might contribute to this poor knowledge and perceptions.

    The SF-6Dv2 is an updated version of the SF-6D, with improved consistency and dimension descriptors, and value sets are required. The aim of this study was to estimate an SF-6Dv2 value set for Australia using an international protocol, while secondary aims were to compare Australian and UK values and to understand heterogeneity.

    The study design was based on the SF-6Dv2 valuation protocol, which involved the administration of two different discrete choice experiment (DCE) tasks. The first task presented pairs of SF-6Dv2 health states with duration (Design 1), and the second task presented triplets that appended ‘immediate death’ to the health state pairs (Design 2). Respondents completed 12 choice sets online (10 from Design 1; 2 from Design 2). Value sets were estimated for Design 1 separately and combining Designs 1 and 2 using a variety of logit model specifications. Value set characteristics were compared descriptively with the UK model.

    The sample included 3001 Australians. A range of ordered and consistent models were estimated. The models only including Design 1 data resulted in a relatively wider utility range. The value range was reduced by an interaction added when the health state included a dimension with a severe level. The model matched with the UK value set resulted in a larger utility range and consistent ordering of the most important dimensions.

Our recent studies suggest that Hsp70-binding protein 1 (HspBP1), an Hsp70/CHIP inhibitor that reduces protein folding, is highly expressed in neuronal cells and accounts for the accumulation of the HD protein huntingtin (HTT) in neuronal cells. To further determine the role of HspBP1 in regulation of mutant protein accumulation, we investigated whether increasing expression of HspBP1 in glial cells can also induce the accumulation of endogenous mutant HTT in glial cells and yield non-cell-autonomous toxic effects. We performed stereotaxic injection of AAV to selectively express HspBP1 in astrocytes in the brains of HD140Q knock-in (KI) mice that express mutant HTT ubiquitously but do not display obvious neurodegeneration. However, HspBP1 expression in HD140Q astrocytes led to the increased accumulation of endogenous mutant HTT and robust neuronal loss in the striatum of HD140Q KI mice. In transgenic HD mice that selectively express mutant HTT in astrocytes, increased accumulation of mutant HTT in astrocytes via HspBP1 expression did not elicit neurodegeneration but could exacerbate neurological symptoms. Consistently, suppressing the expression of endogenous HspBp1 in the striatum of HD140Q KI mice via CRISPR/Cas9 led to a significant reduction of mutant HTT accumulation. Our findings suggest that although endogenous mutant HTT in astrocytes can exacerbate neurological symptoms, it mediates neurodegeneration only when mutant HTT is also accumulated in neuronal cells.

    Since 2010, 10-valent (PCV10) and 13-valent pneumococcal conjugate vaccines (PCV13) have been available as part of infant national immunization programs. Belgium is as one of the few countries that implemented PCV13 (2007-2015), switched to PCV10 (2015-2018) and then switched back to PCV13 (2018-present) after observing increases in disease. We assessed the impacts of both historical and prospective PCV choice in the context of the Belgian health care system and used this experience to validate previously developed economic models.

    Using historical incidence (2007-2018) of pneumococcal disease for Belgian children aged < 16years, observed invasive pneumococcal disease (IPD) trends from surveillance data were used to estimate future disease in a given PCV13- or PCV10-based program. We compared observed incidence data with two modeled scenarios (1) the 2015 switch to PCV10 and (2) a hypothetical continuation of PCV13 in 2015. Finally, we explored the potential impact of PCV choice from 2019 to 2023 by cof PCV13 versus PCV10 in other countries may have underestimated the benefit of PCV13. These results highlight the importance of continually protecting against vaccine-preventable pneumococcal serotypes.There are few studies regarding imaging markers for predicting postoperative rebleeding after stereotactic minimally invasive surgery (MIS) for hypertensive intracerebral haemorrhage (ICH), and little is known about the relationship between satellite sign on computed tomography (CT) scans and postoperative rebleeding after MIS. This study aimed to determine the value of the CT satellite sign in predicting postoperative rebleeding in patients with hypertensive ICH who undergo stereotactic MIS. We retrospectively examined and analysed 105 patients with hypertensive ICH who underwent standard stereotactic MIS for hematoma evacuation within 72 h following admission. Postoperative rebleeding occurred in 14 of 65 (21.5%) patients with the satellite sign on baseline CT, and in 5 of the 40 (12.5%) patients without the satellite sign. This difference was statistically significant. Positive and negative values of the satellite sign for predicting postoperative rebleeding were 21.5% and 87.5%, respectively. Multivariate logistic regression analysis verified that baseline ICH volume and intraventricular rupture were independent predictors of postoperative rebleeding. In conclusion, the satellite sign on baseline CT scans may not predict postoperative rebleeding following stereotactic MIS for hypertensive ICH.

    Regulators have called for greater emphasis on the role of the patient voice to inform medical product development and decision making, and expert guidelines and reports for asthma and chronic obstructive pulmonary disease (COPD) both explicitly recommend the consideration of patient preferences in the management of these diseases. Discrete choice experiments (DCEs) are commonly used to quantify stakeholders’ treatment preferences and estimate the trade-offs they are willing to make between outcomes such as treatment benefits and risks.

    The aim of this systematic literature review is to provide an up-to-date and critical review of DCEs published in asthma and COPD; specifically, we aim to evaluate the subject of preference studies conducted in asthma and COPD, what attributes have been included, stakeholders’ preferences, and the consistency in reporting of instrument development, testing and reporting of results.

    A systematic review of published DCEs on asthma and COPD treatments was conducted using Embase, Medline and the Cochrane Database of Systematic Reviews.
