We pinpoint a previously uninvestigated miRNA in the brain, miR-127, that is highly neuronal, to be associated with increased cell death in the aged, LPS-injected ischemic mice. Target prediction tools indicate that miR-127 interacts with several basally expressed neuronal genes, and of these we verify miR-127 binding to Psmd3. Finally, we report reduced expression of miR-127 in human stroke brains. Our results underline the impact of peripheral inflammation on the outcome of stroke in aged subjects and pinpoint molecular targets for restoring endogenous neuronal capacity to combat ischemic stroke.This paper reports how we applied systems dynamics methods to gain insight into the complexity of obesity-related behaviors in youth, including diet, physical activity, sedentary behavior, and sleep, by integrating a literature review into causal loop diagrams (CLDs). Results showed that the CLDs consisted of multiple subsystems and three types of dynamics appeared, including (1) feedback loops, (2) connections between feedback loops and subsystems, and (3) mechanisms. We observed clear similarities in the dynamics for the four behaviors in that they relate to “traditional” subsystems, such as home and school environments, as well as to newly added subsystems, including macroeconomics, social welfare, and urban systems. The CLDs provided insights that can support the development of intervention strategies, including (1) the confirmation that a range of mechanisms cover and connect multiple levels and settings, meaning that there is no silver bullet to address obesity; (2) understanding of how interventions in one particular setting, such as school, might be influenced by the interactions with other settings, such as urban systems; and (3) a comprehensive view of (un)intended consequences. This way of framing the problem will assist moving towards public health interventions that respond to and operate in the complexity of the real world.A selection of pyrene-based liquid crystal dimers have been prepared, containing either methylene-ether or diether linked spacers of varying length and parity. All the diether linked materials, CBOnO.Py (n=5, 6, 11, 12), exhibit conventional nematic and smectic A phases, with the exception of CBO11O.Py which is exclusively nematic. The methylene-ether linked dimer, CBnO.Py, with an even-membered spacer (n=5) was solely nematogenic, but odd-members (n=6, 8, 10) exhibited both nematic and twist-bend nematic phases. Replacement of the cyanobiphenyl fragment by cyanoterphenyl giving CT6O.Py, gave elevated melting and nematic-isotropic transition temperatures, and SmA and SmCA phases were observed on cooling the nematic phase. The stability of the glassy twist-bend nematic phase allowed for its study using AFM, and the helical pitch length, PTB , was measured as 6.3 and 6.7 nm for CB6O.Py and CB8O.Py, respectively. These values are comparable to the shortest pitch of a twist-bend nematic phase measured to date.This study assesses toxicity of groundwater from remediated fuel spill sites, as the final phase of an environmental risk assessment of contaminated sites at sub-Antarctic Macquarie Island, Tasmania, Australia. To complement previous terrestrial ecotoxicological research, we determine risk to marine environments from residual biodegraded hydrocarbon contaminants in groundwater discharges. Direct toxicity assessments were conducted on 7 composite groundwater test solutions, adjusted to ambient seawater salinity. Eleven native marine invertebrates (from varied taxa gastropods, bivalves, flatworms, amphipods, copepods, isopods) were exposed and observed for up to 21 d. Lethal time estimates (LT10, LT50) showed sensitivity was time dependent (LT10s = 4-15 d) and variable between species. Three species showed no response to any test solution, and most species did not respond for up to 5 d. Data were interpreted using an expert judgment response matrix with multiple lines of evidence to predict risk. No consistent patterns in the relative toxicity of test solutions, based on polar or nonpolar hydrocarbon concentrations, were identified. Although toxicity was observed in some species, this was only under worst-case conditions of undiluted, continuous, extended exposure. Natural dynamics of the site, including low groundwater discharge rates, high rainfall, and a highly energetic receiving environment, ensure groundwater is rapidly diluted and dispersed. In this context, and based on site conditions at the time of testing, these toxicity assessments provide robust evidence that residual contamination in groundwater at remediated sites at Macquarie Island is unlikely to represent a risk to the adjacent marine communities tested. We report on QoL and psychiatric illness diagnoses in a modern cohort of adults with achondroplasia. SF-36 Health Survey scores from adults with achondroplasia were compared to general population scores. Demographics, physical measurements, and psychiatric illness diagnoses were recorded from medical records. The achondroplasia population had lower scores than the general population in all categories. Most people with achondroplasia (56%) had a diagnosed psychiatric illness. Those with a diagnosed psychiatric illness had lower scores in physical functioning, role limitations due to physical and emotional health, and mental health. Pain, energy/fatigue, and general health scale scores were roughly equivalent (15 points higher in individuals with psychiatric illness diagnoses. Adults with achondroplasia report significantly lower physical and mental well-being and had nearly 3× the rate of psychiatric illness diagnosis than the general population, highlighting the importance of total care for this population.
