Tumor xenograft models can create a high capacity to study human tumors and discover efficient therapeutic approaches. Here, we aimed to develop the gamma-radiated immunosuppressed (GIS) mice as a new kind of tumor xenograft model for biomedical studies. First, 144 mice were divided into the control and treated groups exposed by a medical Cobalt-60 apparatus in 3, 4, and 5 Gy based on the system outputs. Then, 144 BALB/c mice were divided into four groups; healthy, xenograft, radiation, and radiation + xenograft groups. The animals in the xenograft and radiation + xenograft groups have subcutaneously received 3 × 106 MCF-7 cells 24 h post-radiation. On 3, 7, 14, and 21 days after cell injection, the animals were sacrificed. Then, the blood samples and the spleen and tumor tissues were removed for the cellular and molecular analyses. The whole-body gamma radiation had a high immunosuppressive effect on the BALB/c mice from 1 to 21 days post-radiation. The macroscopic and histopathological observations have proved that the created clusters’ tumor structure resulted in the xenograft breast tumor. There was a significant increase in tumor size after cell injection until the end of the study. Except for Treg, the spleen level of CD4, CD8, CD19, and Ly6G was significantly decreased in Xen + Rad compared to the Xen alone group on 3 and 7 days. Unlike IL-4 and IL-10, the spleen level of TGF-β, INF-γ, IL-12, and IL-17 was considerably decreased in the Xen + Rad than the Xen alone group on 3 and 7 days. The spleen expressions of the VEGF, Ki67, and Bax/Bcl-2 ratio were dramatically increased in the Xen + Rad group compared to the Xen alone on 3, 7, 14, and 21 days. Our results could confirm a new tumor xenograft model via an efficient immune-suppressive potential of the whole-body gamma radiation in mice.Fear, anxiety, and preference in fish are generally evaluated by video-based behavioural analyses. We previously proposed a system that can measure bioelectrical signals, called ventilatory signals, using a 126-electrode array placed at the bottom of an aquarium and achieved cameraless real-time analysis of motion and ventilation. In this paper, we propose a method to evaluate the emotional state of fish by combining the motion and ventilatory indices obtained with the proposed system. In the experiments, fear/anxiety and appetitive behaviour were induced using alarm pheromone and ethanol, respectively. We also found that the emotional state of the zebrafish can be expressed on the principal component (PC) space extracted from the defined indices. The three emotional states were discriminated using a model-based machine learning method by feeding the PCs. Based on discrimination performed every 5 s, the F-score between the three emotional states were as follows 0.84 for the normal state, 0.76 for the fear/anxiety state, and 0.59 for the appetitive behaviour. These results indicate the effectiveness of combining physiological and motional indices to discriminate the emotional states of zebrafish.The rapid development of the optical-cycle-level ultra-fast laser technologies may break through the bottleneck of the traditional ultra-intense laser [i.e., Petawatt (PW, 1015 W) laser currently] and enable the generation of even higher peak-power/intensity lasers. Herein, we simulate an ultra-broadband concept for the realization of an Exawatt-class (EW, 1018 W) high peak-power laser, where the wide-angle non-collinear optical parametric chirped-pulse amplification (WNOPCPA) is combined with the thin-plate post-compression. A frequency-chirped carrier-envelope-phase stable super-continuum laser is amplified to high-energy in WNOPCPA by pumping with two pump-beamlets and injected into the thin-plate post-compression to generate a sub-optical-cycle high-energy laser pulse. The numerical simulation shows this hybrid concept significantly enhances the gain bandwidth in the high-energy amplifier and the spectral broadening in the post-compression. By using this concept, a study of a prototype design of a 0.5 EW system is presented, and several key challenges are also examined.We have developed two candidate vaccines to protect against multiple strains of Strep A infections. Atglistatin cost The candidates are combinatorial synthetic peptide vaccines composed of a M protein epitope (J8 or p*17) and a non-M protein epitope (K4S2). To enhance immunogenicity, each peptide is conjugated to the carrier protein CRM197 (CRM) and formulated with aluminium hydroxide adjuvant Alhydrogel (Alum) to make the final vaccines, J8-CRM + K4S2-CRM/Alum and p*17-CRM + K4S2-CRM/Alum. The safety and toxicity of each vaccine was assessed. Sprague Dawley rats were administered three intramuscular doses, over a six-week study with a 4-week recovery period. A control group received CRM only formulated with Alum (CRM/Alum). There was no evidence of systemic toxicity in the rats administered either vaccine. There was an associated increase in white blood cell, lymphocyte and monocyte counts, increased adrenal gland weights, adrenocortical hypertrophy, and increased severity of granulomatous inflammation at the sites of injection and the associated inguinal lymph nodes. These changes were considered non-adverse. All rats administered vaccine developed a robust and sustained immunological response. The absence of clinical toxicity and the development of an immunological response in the rats suggests that the vaccines are safe for use in a phase 1 clinical trial in healthy humans.Micronutrient deficiencies and stunting rates are high in many low-income countries. Increasing and diversifying food intake are often challenging for small-scale farmers in lowland areas as flooding often results in crop losses and drowning of livestock. A cluster-randomised controlled trial was conducted over 12-months in Bangladesh, involving 150 small-scale duck rearing households, including 50 control, and 50 households each in two intervention arms. Interventions focussing on improving duck health and duck nutrition were applied on a village level. Data analysis focussed on assessing the impact of interventions on duck mortality, sales and consumption, and on dietary diversity of household members. Improved duck rearing increased the consumption and the sales of ducks. Household selling more ducks were more likely to purchase and consume milk products, contributing to an improved households’ dietary diversity. Our results suggest that improving duck rearing can provide a suitable and sustainable alternative to maintain and improve dietary diversity of households in flood-prone areas.
