001) whereas LLVA was not (p = 0.11). These findings show that LLVA does not respond as well as BCVA following any treatment for PDR, even though BCVA and LLVA both test foveal function.Amyloid aggregation of human ataxin-3 (ATX3) is responsible for spinocerebellar ataxia type 3, which belongs to the class of polyglutamine neurodegenerative disorders. It is widely accepted that the formation of toxic oligomeric species is primarily involved in the onset of the disease. For this reason, to understand the mechanisms underlying toxicity, we expressed both a physiological (ATX3-Q24) and a pathological ATX3 variant (ATX3-Q55) in a simplified cellular model, Escherichia coli. It has been observed that ATX3-Q55 expression induces a higher reduction of the cell growth compared to ATX3-Q24, due to the bacteriostatic effect of the toxic oligomeric species. Furthermore, the Fourier transform infrared microspectroscopy investigation, supported by multivariate analysis, made it possible to monitor protein aggregation and the induced cell perturbations in intact cells. In particular, it has been found that the toxic oligomeric species associated with the expression of ATX3-Q55 are responsible for the main spectral changes, ascribable mainly to the cell envelope modifications. A structural alteration of the membrane detected through electron microscopy analysis in the strain expressing the pathological form supports the spectroscopic results.Human papillomavirus 16 (HPV16) exhibits a strong oncogenic potential mainly in cervical, anogenital and oropharyngeal cancers. The E6 and E7 viral oncoproteins, acting via specific interactions with host cellular targets, are required for cell transformation and maintenance of the transformed phenotype as well. We previously demonstrated that HPV16E7 interacts with the actin-binding protein gelsolin, involved in cytoskeletal F-actin dynamics. Herein, we provide evidence that the E7/gelsolin interaction promotes the cytoskeleton rearrangement leading to epithelial-mesenchymal transition-linked morphological and transcriptional changes. E7-mediated cytoskeletal actin remodeling induces the HIPPO pathway by promoting the cytoplasmic retention of inactive P-YAP. These results suggest that YAP could play a role in the “de-differentiation” process underlying the acquisition of a more aggressive phenotype in HPV16-transformed cells. A deeper comprehension of the multifaceted mechanisms elicited by the HPV infection is vital for providing novel strategies to block the biological and clinical features of virus-related cancers. In December 2019, the emergence of SARS-CoV-2 virus in China led to a pandemic. Since both Influenza Like Illness (ILI) and COVID-19 case definitions overlap, we re-investigated the ILI cases using PCR for the presence of SARS-CoV-2 in 739 nasopharyngeal swabs collected from November 2019 to March 2020. SARS-CoV-2 RNA was found in 37 samples (5%) collected mostly during February 2020. It was followed by confirmation of evolutionary and spatial relationships using next generation sequencing (NGS). We observed that the overall incidence of ILI cases during 2019-2020 influenza season was considerably higher than previous years and was gradually replaced with SARS-CoV-2, which indicated a silent transmission among ambulatory patients. Sequencing of representative isolates confirmed independent introductions and silent transmission earlier than previously thought. Evolutionary and spatial analyses revealed clustering in the GH clade, characterized by three amino acid substitutions in spike gene (D614G), RdRp (P323L) and NS3 (Q57H). P323L causes conformational change near nsp8 binding site that might affect virus replication and transcription. In conclusion, assessment of the community transmission among patients with mild COVID-19 illness, particularly those without epidemiological link for acquiring the virus, is of utmost importance to guide policy makers to optimize public health interventions. The detection of SARS-CoV-2 in ILI cases shows the importance of ILI surveillance systems and warrants its further strengthening to mitigate the ongoing transmission of SARS-CoV-2. The effect of NS3 substitutions on oligomerization or membrane channel function (intra- and extracellular) needs functional validation.Microalgae separation technology is essential for both executing laboratory-based fundamental studies and ensuring the quality of the final algal products. PJ34 manufacturer However, the conventional microalgae separation technology of micropipetting requires highly skilled operators and several months of repeated separation to obtain a microalgal single strain. This study therefore aimed at utilizing microfluidic cell sorting technology for the simple and effective separation of microalgae. Microalgae are characterized by their various morphologies with a wide range of sizes. In this study, a contraction-expansion array microchannel, which utilizes these unique properties of microalgae, was specifically employed for the size-based separation of microalgae. At Reynolds number of 9, two model algal cells, Chlorella vulgaris (C. vulgaris) and Haematococcus pluvialis (H. pluvialis), were successfully separated without showing any sign of cell damage, yielding a purity of 97.9% for C. vulgaris and 94.9% for H. pluvialis. The result supported that the inertia-based separation technology could be a powerful alternative to the labor-intensive and time-consuming conventional microalgae separation technologies.Timely and accurate crop growth monitoring and yield estimation are important for field management. The traditional sampling method used for estimation of ramie yield is destructive. Thus, this study proposed a new method for estimating ramie yield based on field phenotypic data obtained from unmanned aerial vehicle (UAV) images. A UAV platform carrying RGB cameras was employed to collect ramie canopy images during the whole growth period. The vegetation indices (VIs), plant number, and plant height were extracted from UAV-based images, and then, these data were incorporated to establish yield estimation model. Among all of the UAV-based image data, we found that the structure features (plant number and plant height) could better reflect the ramie yield than the spectral features, and in structure features, the plant number was found to be the most useful index to monitor the yield, with a correlation coefficient of 0.6. By fusing multiple characteristic parameters, the yield estimation model based on the multiple linear regression was obviously more accurate than the stepwise linear regression model, with a determination coefficient of 0.
