The unavailability of virus-specific drugs has created an opportunity to identify potential therapeutic agents that can control the rapid transmission of this pandemic. Here, the mechanisms of the inhibition of the RNA-dependent RNA polymerase (RdRp), responsible for the replication of the virus in host cells, are examined by different ligands, such as Remdesivir (RDV), Remdesivir monophosphate (RMP), and several artificially expanded genetic information systems (AEGISs) including their different sequences by employing molecular docking, MD simulations, and MM/GBSA techniques. It is found that the binding of RDV to RdRp may block the RNA binding site. However, RMP would acquire a partially flipped conformation and may allow the viral RNA to enter into the binding site. The internal dynamics of RNA and RdRp may help RMP to regain its original position, where it may inhibit the RNA-chain elongation reaction. Remarkably, AEGISs are found to obstruct the binding site of RNA. It is shown that dPdZ, a two-nucleotide sequence containing P and Z would bind to RdRp very strongly and may occupy the positions of two nucleotides in the RNA strand, thereby denying access of the substrate-binding site to the viral RNA. Thus, it is proposed that the AEGISs may act as novel therapeutic candidates against the SARS-CoV-2. In this study, we elucidate the therapeutic effects and mechanistic basis of Adhatoda vasica (AV) aqueous extract on mouse models of acute allergic as well as severe asthma subtypes at physiological, histopathological, and molecular levels. Oral administration of AV extract attenuates the increased airway resistance and inflammation in acute allergic asthmatic mice and alleviates the molecular signatures of steroid (dexamethasone) resistance like IL-17A, KC (murine IL-8 homologue), and HIF-1α (hypoxia-inducible factor-1α) in severe asthmatic mice. AV inhibits HIF-1α levels through restoration of expression of its negative regulator-PHD2 (prolyl hydroxylase domain-2). This qualitative study investigated organizational and community impact of Stewards of Children (SOC) in the context of a statewide initiative to prevent child sexual abuse. Participants, representing 11 sites, were SOC trainers or in administrative and leadership roles with their organizations. The study focused on four research questions 1) How has your organization been impacted (by SOC), if at all? 2) How have your relationships with other organizations and groups changed, if at all? 3) How has the perception of your organization changed in the community, if at all? and 4) Have you noticed changes in your community as a result of Stewards of Children trainings? The findings include three themes and seven subthemes. Participants uniformly agreed that SOC had an impact on their organization, their relationship with partner and new-partner organizations, and that it contributed to increased awareness about a) the problem of child sexual abuse overall, b) their organization among multiple groups and partners, and c) the full array of services and expertise provided by SOC organizations.Gottfredson and Hirschi’s General Theory of Crime’s (GTC) claim to explain crime, at all times, and in all places, has received considerable empirical support using Western, Scandinavian, and Asian samples. However, the theory is yet to be tested using a Nigerian sample. Using cross-sectional data from a sample of 1,192 junior secondary school (middle school) students in Nigeria, this study examines the explanatory power of Gottfredson and Hirschi’s GTC in predicting bullying perpetration and victimization in that context and culture. Binomial logistic regression was used to test the hypothesis that the bullying perpetration and victimization experiences of these students would be negatively related to self-control. Study provides support for the cross-cultural potency of the GTC as low self-control predicted the bullying and victimization experiences of these students regardless of gender. Students’ perception of a negative school environment was also a significant predictor of bullying. This is the first test of the GTC with a Nigerian sample and hopefully, will be a springboard for effective bullying prevention policy/programs in Nigerian schools.
