The combination of both EI and CI mass spectrum analysis improves molecular identification, thanks to the detection of molecular, deprotonated or protonated ion of highest intensity and fragment formula assignments. The EI and NCI NH3 combination allows formula assignments up to 94% of our database with limit of detection up to 7 ppm. This procedure has been validated for untargeted GC-FT-Orbitrap-MS analysis of VOC coming from the processing of cometary ice analogs.The synergistic effect of chiral ionic liquids composed of tetraalkylammonium ions and the amino acids Asn, Asp or Pro on the enantioseparations of dipeptides mediated by β-cyclodextrin and 2-hydroxypropyl-β-cyclodextrin in capillary electrophoresis was studied. Addition of a chiral ionic liquid resulted in a concentration-dependent increase in the enantioresolutions compared to the sole presence of a cyclodextrin in the background electrolyte. The extent varied with the tetraalkylammonium cation (tetramethylammonium versus tetrabutylammonium) as well as the amino acid component of the ionic liquid. The presence of a chiral ionic liquid did not counteract the pH-dependent reversal of the enantiomer migration order of the dipeptides Ala-Phe, Ala-Tyr and Phe-Phe when increasing the pH of the background electrolyte from 2.5 to 3.5. Comparing the effect of a chiral ionic liquid based on Asp with the addition of equimolar concentrations of the individual components of the ionic liquid, a diverse picture was observed. In some cases, higher resolution values were obtained with the chiral ionic liquid, while for other cases superior enantioseparations were obtained upon separate addition of the amino acid component and a tetraalkylammonium chloride. With regard to the stereochemistry of the amino acid, a superior effect was typically observed using the l-configured amino acid, but in some cases higher resolution values were found in the presence of d-Asp. The rationale for the diverse observations is not obvious and may be due to the zwitterionic nature of analytes as well as the amino acid component of the chiral ionic liquid.Comprehensive elucidation of the composition of multiprotein complexes in model organisms is essential to understand conserved biological systems, but large-scale mapping physical association networks is still challenging due to limited throughput of present methods. In this work, a strategy coupling array-based online two-dimensional liquid chromatography (array-based 2D-LC) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) was demonstrated for high throughput and in-depth identification of protein complexes from cultured human HeLa cell extracts. Mixed-bed ion-exchange column was employed as the first dimensional (1stD) separating mode and an array consisting of eight reversed phase columns was developed as the second dimensional (2ndD) mode. Taking advantage of array parallel strategy, this online system showed an 8-fold increase in throughput. After array-based online 2D-LC separation, altogether 256 × 2ndD fractions were collected for further LC-MS/MS analysis. Public databases of protein-protein interaction (PPI) and co-elution curves identified by LC-MS were applied to reconstruct the protein complexes. A rigorous inspection was operated by cataloging the protein complexes into chromatographic fractions to minimize the number of false positives. As result, a total number of 4,436 proteins were identified and 26,092 elution curves were graphed. A network consisting of 47,745 PPIs was established among 2,201 proteins and presented 1,530 putative protein complexes with high confidence. Most of the identified PPIs were linked to diverse biological processes and may reveal further disease mechanism and therapeutic strategy.
Patients seeking treatment for their asthma are most likely motivated by a change in their experience of symptoms, but primary complaints are not always related to the pulmonary system. This study aimed to determine the frequency of such extrapulmonary symptoms in asthma outpatients and their association with psychopathology and asthma outcomes.
This cross-sectional study utilized data collected as part of a nationwide, clinical-epidemiological study. The final sample of 572 asthma patients represented all levels of asthma control and severity. Information on demographics and respiratory function was obtained from physicians’ documentation. Symptoms were explored using a standardized checklist.
Primary symptoms reported by asthma patients were not necessarily airway-related. Patients reported feeling at least occasionally “tired” (72.1%) and “exhausted” (66.8%) more than any other asthma symptom. Surveillance of extrapulmonary symptoms alongside pulmonary function is warranted for an integrated medicine approach to asthma management.
The literature on childhood-onset depression and future compromised vascular function is suggestive but limited. The objective of this study was to determine if arterial stiffness, a predictor of future cardiovascular disease (CVD), measured in young adulthood, is associated with childhood-onset depression.
Cardiometabolic risk factors and pulse wave velocity (PWV), a measure of arterial stiffness, were cross-sectionally assessed in young adults with a history of childhood-onset depression (clinical diagnosis of major depressive episode or dysthymic disorder; N=294 probands; initially recruited via child mental health facilities across Hungary; mean age of first depressive episode=10.4years), their never-depressed full biological siblings (N=269), and never-depressed controls (N=169). The mean ages of probands, siblings, and controls at the PWV visit were 25.6, 25.0, and 21.7years, respectively, and 8.8% of the probands were in a current depressive episode.
Controlling for age, sex, age*sex, education, dhood-onset depression suggest that pediatric depression may predispose to increased CVD risk later in life and warrants further investigation.
